The primary focus of the research is to explore and decipher the mechanisms of extracellular vesicle (EV) delivery at the molecular level.
Key Responsibilities: Develop and optimize novel cell-based assays to study extracellular vesicle delivery. Perform high-throughput CRISPR-Cas9-based genetic screenings to identify key genes and pathways involved in EV delivery. Analyze and interpret large-scale genetic and molecular datasets to uncover new insights into the molecular underpinnings of EV function. Collaborate with a multidisciplinary team to integrate findings with broader research goals.

L'objectif de la thèse sera de mettre au point des biopuces nano/microstructurées à résonance des plasmons de surface pour détecter, caractériser puis doser un ensemble de cibles biologiques multi-échelles (de la molécule à la vésicule extracellulaire) simultanément (format multiplex) au sein de sécrétomes que le/la candidat.e aura contribué à générer.

Extracellular vesicles associated to indole-producing synthetic gut community: a signal pathway between gut microbiota and brain in newborns?
The main objective of this study aims at investigating the BEVs from indole-producing bacteria frequently found in newborn gut microbiota. The study will include the analyses of samples for BEVs production from in vitro bacterial cultures and in vivo experimentations on preclinical models in controlled conditions. BEVs will be characterized by physical and biological assessments after extraction and purification. Biological functions of the BEVs derived from indole- producing bacteria and synthetic gut communities will be determined using our expertise on specific stem cell lines and ex vivo electrophysiological analyses on brain slides.
Candidate profile and required skills: We are looking for a rigorous candidate with creative and collaborative spirit. PhD in Microbiology, Experience/Knowledge in EVs, Scientific English speaking and writing are required. Knowledge in microbiota is recommended. We are looking for a post-doctoral researcher whose objective will be to characterize and determine the role(s) of sEVs produced in the human alveolar epithelium in SARS-CoV-2 infection . The candidate will be in charge of the purification, characterization of sEVs and the infection of cells by different variants of SARS-CoV-2 in the presence or absence of sEVs. The work will be performed between the TRePCa laboratory and the IMRB team 18 (same site). The TRePCa laboratory is located at the Faculty of Health of Créteil and works on the field of extracellular vesicles in different contexts. Team 18 of the IMRB is specialized in the study of respiratory viruses. The candidate will be supervised by Dr Virginie Firlej and Dr Patrice Bruscella The candidate must be highly motivated, preferably with a PhD in cell biology. Skills in biochemistry and molecular biology are required (Western-blot, RT-qPCR, microscopy…). Knowledge in the field of extracellular vesicles and/or virology will be appreciated. The postdoc will carry out independent research on extracellular vesicles (EVs) derived from mesenchymal stromal cells (MSCs) and their functional role in aging. Based on a selection of proteins differentially expressed in EVs from senescent MSCs, the first objective is to validate the selected proteins using functional assays in vitro and possibly in vivo using murine models of arthritis. The secondary objective is to improve the potency of MSCs-derived EVs by modulating the expression of relevant proteins with the aim of enhancing their therapeutic efficacy in osteoarthritis.

Influence of mammary tumor extracellular vesicles in the development of brain metastases.
A 2-year postdoctoral position in extracellular vesicle-dependent cell communication in cancer is available in PRISM laboratory, Inserm UMR-1192, Université de Lille, directed by Pr. Michel Salzet. The postdoc will study the physiopathological features of breast cancer leading to brain metastasis. For this purpose, the extracellular vesicles from in vitro models of tumor microenvironment will be isolated and molecularly characterized. Their potential to prepare metastatic niches and regulate the inflammatory balance will be deciphered. The applicant should have a strong background in cell biology and EV studies including their isolation, labeling and possible in vivo tracking. The postdoc will work on mechanisms of extracellular vesicle release and biodistribution in the context of cardiovascular disease development.
